چکیده
Abstract Background Hepatocellular carcinoma (HCC) is the leading cause of tumor-related death worldwide due to high morbidity and mortality, yet lacking effective biomarkers therapies. Circular RNAs (circRNAs) are a group non-coding that regulate gene expression through interacting with miRNAs, implicating in tumorigenesis progression. A novel circRNA, circTP63, was reported be an oncogene HCC. However, its role HCC remains unclear. Methods qRT-PCR used assess mRNA levels CircTP63 90 pairs tumor adjacent normal tissues from patients, one human hepatic epithelial cell line lines. CCK-8, colony formation, transwell, flow cytometry assays were performed detect cellular function circTP63/miR-155-5p/ZBTB18 cells. xenograft mice models established vivo effect circTP63. Bioinformatic analysis, RNA pull-down luciferase determine interaction among circTP63/miR-155-5p/ZBTB18. Results circTP63 significantly upregulated High closely associated stages, lymph node metastasis, poor prognosis patients. Functionally, knockdown inhibited proliferation, migration, invasion, promoted apoptosis Meanwhile, overexpression enhanced Mechanically, sponge miR-155-5p facilitate ZBTB18 expression, negatively survival rate Furthermore, rescued revealed reduced tumor-promoting on cells induced by can reversed inhibitor or overexpression. Conclusion Our data highlight critical circTP63-miR-155-5p-ZBTB18 regulatory network involved progression, gaining mechanistic insights into circRNAs providing therapeutic targets for treatment.

miR-182-5p promotes hepatocellular carcinoma progression by repressing FOXO3a
BACKGROUND High frequency of recurrence is the major cause of the poor outcomes for patients with hepatocellular carcinoma (HCC). microRNA (miR)-182-5p emerged as a high-priority miRNA in HCC and was found to be related to HCC metastasis. Whether the expression of miR-182-5p in tumor tissue correlated with early recurrence in HCC patients underwent curative surgery was unknown. METHODS Real-t...
متن کاملMicroRNA‐155‐5p promotes hepatocellular carcinoma progression by suppressing PTEN through the PI3K/Akt pathway
MicroRNA-155-5p (miR-155-5p) has been reported to play an oncogenic role in different human malignancies; however, its role in hepatocellular carcinoma (HCC) progression is not clearly understood. In this study, we used real-time PCR in 20 rats with chemically-induced HCC, 28 human HCC tissues, and the matched paracarcinoma tissues, and HCC cell lines to determine the expression patterns of miR...
متن کاملmiR-92a promotes hepatocellular carcinoma cells proliferation and invasion by FOXA2 targeting
Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...
متن کاملLong non-coding RNA MIAT promotes breast cancer progression and functions as ceRNA to regulate DUSP7 expression by sponging miR-155-5p
Long non-coding RNAs (lncRNA) have been reported as key regulators in the progression and metastasis of breast cancer. In this study, we found that the lncRNA myocardial infarction associated transcript (MIAT) expression was upregulated in breast cancer in The Cancer Genome Atlas (TCGA) data sets. We validated that MIAT was higher in breast cancer cell lines and advanced breast tumors than in n...
متن کاملUp-regulated miR-155-5p promotes cell proliferation, invasion and metastasis in colorectal carcinoma.
BACKGROUND AND OBJECTIVE Emerging evidences indicate that miR-155-5p is associated with some cancer tumorigenesis, but their specific effects on proliferation, invasion and metastasis of colorectal carcinoma (CRC) are still poorly understood. The aim of the study is to investigate miR-155-5p effect on proliferation and invasion metastasis of CRC. METHODS Retrospectively analyzed clinicopathol...
